Abstract

Background: Antibiotics are generally used to treat infections by bacteria, but the use of antibiotics can cause side effects such as causing bacteria to become resistant so that the use of antibiotics will be ineffective. Alternative medicine continues to be sought and developed to deal with resistant bacteria. Mole crab shell (Emerita sp.) contains chitosan compounds. Chitosan can provide antibacterial activity. Antibacterial activity test can use the paper disk method. Purpose: Knowing the antibacterial activity test of mole crab shell (Emerita sp.) against Escherichia coli and Staphylococcus aureus bacteria and knowing the comparison of antibacterial activity against Escherichia coli and Staphylococcus aureus bacteria. Methode: The method of making chitosan is carried out through the stages of demineralization, deproteination, and deacetylation. Antibacterial activity test was carried out using the paper disk method. Result: In the manufacture of chitosan, the deacetylation degree value was 57.62%. The results of the diameter of the chitosan inhibition zone from mole crab (Emerita sp.) shells for Escherichia coli bacteria with chitosan concentrations of 50 ppm, 100 ppm, 150 ppm, 200 ppm, 250 ppm were 3.36 mm; 4.66mm; 5.86mm; 6.78mm; 10 mm while for Staphylococcus aureus bacteria, which is 0.34 mm; 2.31mm; 4.08mm; 6.08mm; 8.46mm. Conclusion: Chitosan from mole crab shells (Emerita sp.) has antibacterial activity against Escherichia coli and Staphylococcus aureus bacteria. The antibacterial activity of chitosan from mole crab (Emerita sp.) shells against Escherichia coli bacteria provided better antibacterial activity compared to Staphylococcus aureus bacteria seen from its inhibition zone. Recommendation: Further research is needed to be developed into preparations as an antibacterial.
