Abstract
Background: Pain is an unpleasant sensory and emotional experience associated with tissue damage. Black turmeric (Curcuma caesia Roxb.) has analgesic activity with fewer side effects compared to conventional drugs. Objective: To determine the activity and optimal dose of aqueous extract of black turmeric as an analgesic in male white rats using the hotplate test. Method: This experimental laboratory study involved infusing black turmeric rhizomes and using 25 male white rats as subjects. The rats were divided into five treatment groups: normal kontrol, positif kontrol, negatif kontrol, and groups receiving black turmeric aqueous extract at doses of 100 mg/kg body weight, 200 mg/kg body weight, and 400 mg/kg body weight. The hotplate test model was used to assess pain response. The treatments were administered orally for 5 days, and pain response was measured using the % protection with data analyzed using SPSS version 25. Results: The black turmeric extract (Curcuma caesia Roxb.) demonstrated analgesic activity, as indicated by the mean difference in hotplate response for the extract doses of 100 mg/kg body weight, 200 mg/kg body weight, and 400 mg/kg body weight, which were 12.26, 11.30, and 13.15, respectively, with a significance value of <0.05. Conclusion: Black turmeric rhizome has an analgesic effect on heat-induced pain in the hotplate test at a dose of 400 mg/kg body weight. Recommendation: It is suggested to conduct further tests on the levels of flavonoids, alkaloids, tannins, phenols, and saponins in the aqueous extract of black turmeric to determine the specific compounds responsible for the analgesic activity.
