Abstract

Background: Diabetes mellitus is a metabolic disorder characterized by decreased insulin production by the pancreas. In Indonesia, the prevalence of diabetes was 6.2% in 2020, and this number is expected to rise by 2045. Black turmeric is one of the natural substances that can be used as an alternative treatment for diabetes. Objective: To determine the activity and optimal dose of black turmeric rhizome infusion as an antidiabetic treatment in streptozotocin-induced rats. Method: This laboratory experimental study used black turmeric rhizome infusion on 24 male Wistar rats. The rats were divided into 6 treatment groups: normal control, positive control, negative control, and groups receiving black turmeric rhizome infusion at doses of 5%, 10%, and 15%. The study employed a diabetes model induced with streptozotocin at 45 mg/kg body weight via intraperitoneal injection. Treatment was administered orally for 21 days, with blood glucose levels measured on days 7, 14, and 21 using a glucometer. Blood glucose reduction was calculated using the percentage reduction formula and analyzed with SPSS version 25. Results: SPSS analysis showed average reductions of -53.250 for the 5% infusion, -66.000 for the 10% infusion, and -75.500 for the 15% infusion. The p-values were 0.083, 0.049, and 0.030, respectively. Conclusion: Black turmeric rhizome has antidiabetic effects, with the most effective dose being 15%. Recommendation: Further research is needed on in vitro antidiabetic testing and toxicity evaluation of black turmeric rhizome infusion.
