Abstract

Background: Fluoride is a component in toothpaste that prevents caries and strengthens tooth enamel. Herbal toothpastes are increasingly in demand as a natural alternative, but still require fluoride content within safe limits. Excess fluoride can cause dental to skeletal fluorosis. The Indonesian National Standard sets the safe limit of fluoride levels in toothpaste at 800-1500 ppm. Validation of the analysis method of fluoride content in herbal toothpaste is important to ensure product safety and quality. Purpose: To determine the validity of UV-Vis spectrophotometric method with SPADNS-zirconyl acid reagent in determining fluoride levels in herbal toothpaste (PG) and its compliance with SNI. Methods: This research is a laboratory experimental study with UV-Vis spectrophotometric method validation. Parameters validated include specificity, linearity, precision, accuracy, LOD, and LOQ. Samples consisted of five types of herbal toothpaste. Results: The UV-Vis spectrophotometric method proved valid for all parameters. Specificity showed the absence of matrix interference. Linearity was shown by a correlation coefficient of 0.9988. Precision was qualified with %RSD of 1.0332%, and accuracy had a %recovery of 102.20%. The LOD value was 5.76 ppm and LOQ was 17.44 ppm. Analysis of five herbal toothpaste samples showed PG1 (802.35 ppm) and PG2 (910.97 ppm) met SNI standards, PG3 (5000 ppm) and PG4 (4810.81 ppm) exceeded the limit, while PG5 did not contain fluoride according to package claims. Conclusion: UV-Vis spectrophotometric method with SPADNS-zirconyl acid reagent is valid for determining fluoride content in herbal toothpaste, but not all samples meet SNI standards. Recommendation: Optimization of analysis time, addition of sample variations, and utilization of comparative methods.
