Abstract

Background: The 96% ethanolic extract of nut grass tuber (Cyperus rotundus L.) has repellent activity due to its flavonoid content, thus has the potential to be developed into a mosquito repellent lotion. Objective: To determine the optimal formula for 96% ethanol extract lotion of rumput teki (Cyperus rotundus l.) tubers using the simplex lattice design method. Methods: Formulation optimization was carried out using the Simplex Lattice Design (SLD) method by varying the concentrations of Tween 80, Span 80, and Cetyl Alcohol with response parameters including viscosity, spreadability, and adhesiveness. The optimal formula was validated using a T-test. Evaluation included organoleptic properties, homogeneity, spreadability, adhesiveness, pH, emulsion type, physical stability, and centrifugation stability. Results: The optimization results show the optimum formula with a composition of Tween 80 5.9%, Span 80 1%, and Cetyl Alcohol 3% with a desirability value of 0.915. This formula produces a viscosity of 6.444±277.8; adhesion time of 6 minutes 22 seconds; spreadability of 6.24 cm±0.135, with a pH of 6.67±0.100. The One Sample T-test shows that not all responses meet the p>0.05 criteria, indicating that the experimental results have a significant difference from the SLD prediction. The evaluation of the optimal formula yields a semi-solid lotion, brown in color, with a distinct chocolate fragrance. Conclusion: The optimized formulation of lotion containing 5.9% Tween 80, 1% Span 80, and 3% Cetyl Alcohol meets quality requirements. Recommendation: Further studies are needed on irritation testing, toxicity testing, and antioxidant activity of the formulation.
