Abstract
Background: Flavonoids are secondary metabolites that have many benefits as an alternative treatment. Taro (Colocasia esculenta L.) is a plant that contains flavonoids. Objectives: To determine the total levels of flavonoids contain in extract and fractions of taro leaves (Colocasia esculenta L.) using the Uv-Vis Spectrophotometry method. Methods: This research is experimental using taro leaves (Colocasia esculenta L.). Taro leaves were extracted by maceration using 96% ethanol solvent. The extract was standardized with 2 parameters, namely specific and non-specific. Specific parameters include organoleptic test, water soluble extract content and ethanol soluble extract content. Non-specific parameters include drying shrinkage test, water content test, total ash content test and acid insoluble ash content test. The extract was fractionated with n-hexane, ethyl-acetate and methanol . Extracts and fractions of taro leaves (Colocasia esculenta L.) were tested in tubes and TLC. Determination of total flavonoid content was carried out by Uv-Vis spectrophotometry. Result: Viscous extract, water soluble extraxt content was 8,8%, ethanol soluble extract content was 27,2%. Drying shrinkage 3,4%, moisture content 2,55, total ash content 2,0% and acid insoluble ash content 0,5%. Ethanol extract 96%, ethil-acetat fraction and methanol fraction positively contain flavonoids. Thetotal flavonoid content of the ethanol extract 96%, n-hexane fraction, ethil-acetatce fraction and methanol fraction of each was 10,27 mgQE/g, 0,136 mgQE/g, 8,85 mgQE/g and 8,15 mgQE/g. Conclusion: The highest total flavonoid content was obtained from 96% ethanol extract, which was 10.27 mgQE/g). Recommendation: Tests for the levels of secondary metabolites such as tannins, saponins, alkaloids and steroids were carried out to obtain a more varied comparison of data.
