Abstract
Background: Diarrhea is one of the main digestive diseases in Indonesia. Escherecia coli bacteria is one of the main pathogens that cause diarrhea. Papaya seeds are part of the papaya plant, which has antibacterial properties that cause diarrhea. Objective: To determine the concentration of papaya seed extract (Carica papaya L.), which can inhibit the growth of Escherecia coli bacteria. Method: This research uses experimental method using the soxhlet extraction technique. The obtained extract was subjected to tube tests. The total terpenoid content of the extract was then determined using UV-Vis spectrophotometry. Following this, antibacterial activity was tested using the disk diffusion method at concentrations of 3.125%, 6.25%, 12.5%, 25%, 50%, and 100% to evaluate the inhibition of Escherichia coli growth. The results were subsequently analyzed using One-Way Anova. Results: According to this research, no inhibition zone forms at concentrations of 3.125, 6.25, 12.5, and 25%, indicating no significant difference. There were significant differences between concentrations of 50 and 100% (P>0,05), while the positive and negative controls also had significant differences compared to all extract concentrations (p<0.05). Conclusion: Concentrations of 50% and 100% showed very strong inhibition effectiveness with inhibition zones of 24.4 mm and 31.1 mm, respectively. However, at concentrations of 3.125%, 6.25%, 12.5%, and 25%, no inhibition zones were formed. Recommendation: To get better results, factors in each process that can influence research results must be considered.
