Abstract

Background : Diabetes mellitus is a disorder of carbohydrate, fat and protein metabolism which is characterized by an increase in body blood sugar levels or hyperglycemia. Administration of oral antidiabetic drugs is one of the pharmacological therapies given to diabetics, but the long-term use of oral antidiabetic drugs can cause side effects and require quite expensive costs.plant (Elaeocarpus ganitrus Roxb.) is one of the natural ingredients that can be used as an alternative treatment for diabetes mellitus with minimal side effects. Objective: This study aimed to determine the effect and effective dose of ethanolic extract of ganitri leaf (Elaeocarpus ganitrus Roxb.) as antidiabetic in streptozotocininduced rats. Method : This research is a laboratory experimental study using ethanol extract of ganitri leaves (Elaeocarpus ganitrus Roxb.) with the research subjects of 25 male white rats of wistar strain (Rattus norvegicus). Test animals were divided into 5 treatment groups, namely negative control (Na-CMC 0.5%), positive control (Amaryl tablet 0.04 mg/kgBW), group of ethanol extract of ganitri leaves at doses of 50, 100 and 200 mg/kgBW. This study used a diabetic rat model that was induced using a streptozotocin 40 mg/kgBW intra peritoneally. The treatment was given orally for 21 days and blood sugar levels were measured every 7 days, namely on days 7, 14 and 21 using a glucometer. Blood samples were taken through the injured tail vein. The decrease in blood sugar levels was calculated using the % decrease formula and analyzed using the SPSS version 16 program with One Way Anova and Post Hoc-LSD statistical tests. Result : The administration of ethanol extract of ganitri leaves (Elaeocarpus ganitrus Roxb.) at doses of 50, 100 and 200 mg/kgBW with a percentage reduction of 6%, 28%, 53% significantly reduced blood glucose levels in streptozotocin with an  < 0.05. Conclusion : The ethanol extract of ganitri leaves (Elaeocarpus ganitrus Roxb.) had a significant antidiabetic effect on streptozotocin with an effective dose of 200 mg/kgBW ( < 0.05). Recommendation : It is necessary to do further research related to in vitro test and toxicity test of ethanol extract of ganitri leaf.
