Abstract

Background: The flavonoid content in ganitri leaves (Elaeocarpus ganitrus Roxb.) which has antioxidant properties is easily disturbed by temperature and light. In addition, limited absorption in the gastrointestinal and intestinal tract and low solubility in water cause flavonoids to have low bioavailability. Modification of the drug delivery system as a solution to increase the stability and bioavailability of flavonoids in the body needs to be done, one of which is by delivering nanoparticle drugs. Research Objectives: This study aims to determine the influence of sodium alginate and calcium chloride as cross-linking agents on the physical characteristics of ganitri leaf extract nanoparticles (Elaeocarpus ganitrus Roxb.), as well as to determine the optimal ratio of sodium alginate and calcium chloride that can produce ganitri leaf extract nanoparticles (Elaeocarpus ganitrus Roxb.) by the Simplex Lattice Design method. Research Methods: This nanoparticle research was made by ionic gelation method with a comparison of the use of 0.1% sodium alginate and 0.01% calcium chloride. Preparation evaluation includes pH, transmittance, and adsorption efficiency tests as optimization parameters, particle size tests, particle size distribution, polydispersity index and zeta potential tests are carried out on the optimal formula. Test the validity by using t-test to validate the equation of the optimization results. Research Results: The results of Design Expert 13.0.5.0 show that the optimal formula with a ratio of 0.1% sodium alginate and 0.01% calcium chloride 6:4 is used. The prediction results obtained were pH 6.743, transmittance 93.806%, adsorption efficiency 92.056% with a desirabillity value of 1,000. The results of the One Sample T-test all responses were p>0.05, which shows that the Design Expert prediction results are not significantly different from the experimental results. Based on the test results above, it shows that the optimization equation for each parameter is valid. The optimal formula has an average particle size of 256,3nm±32,3112, a polydispersity index of 0,384±0,2879, and a potential zeta of 42,8mV±29,0677. Conclusion: Ganitri leaf extract (Elaeocarpus ganitrus Roxb.) can be formulated as a nanoparticle preparation. The combination of sodium alginate and calcium chloride can have an effect on increased transmittance, adsorption efficiency, and decreased pH. Recommendation: Further research is needed on the manufacture of ethanol-aquadest extract nanoparticles of ganitri leaves using different cross-linking methods and agents.
