Abstract

Background: Diabetes mellitus is a metabolic disorder affecting carbohydrates, proteins, and fats, characterized by hyperglycemia or an increase in blood glucose levels. The hyperglycemia experienced by individuals with diabetes mellitus can trigger oxidative stress, caused by the formation of Reactive Oxygen Species (ROS) or free radicals. These free radicals contribute to a decrease in insulin production as they can cause tissue damage in the pancreatic β-cells. Plants with potential antidiabetic properties include Arumanis mango leaves and guava leaves Objective: To determine the effective dose of a combination of ethanol extracts from Arumanis mango leaves (Mangifera indica L.) and guava leaves (Psidium guajava L.) that exhibits antidiabetic activity in male white rats induced with streptozotocin Methode: This research is experimental in nature, using 32 test animals (Rattus norvegicus) divided into 8 test groups: a healthy control group, a positive control group, a negative control group, Group I (EEDMA 60 mg/200g BW), Group II (EEDJB 50 mg/200g BW), Group III (EEDMA 45 mg/200g BW + EEDJB 12.5 mg/200g BW), Group IV (EEDMA 30 mg/200g BW + EEDJB 25 mg/200g BW), and Group V (EEDMA 15 mg/200g BW + EEDJB 37.5 mg/200g BW). The test animals were induced with diabetes mellitus using streptozotocin at 45 mg/kg BW via intraperitoneal injection, and blood glucose levels were measured using a glucometer every 7 days over 21 days Results: Based on the research results, it shows that with a dose of EEDMA 60 mg/200gBB, EEDJB 50 mg/200gBB, EEDMA 45 mg/200gBB + EEDJB 12.5 mg/200gBB, EEDMA 30 mg/200gBB + EEDJB 25 mg/200gBB, and EEDMA 15 mg/ 200gBW + EEDJB 37.5 mg/200gBW the results of successive reductions in blood sugar levels are 39%, 34%, 52%, 42% and 43%. The results of the Howell Games statistical test with p values are 0.280, 0.374, 0.140, 0.231, 0.225 respectively. Conclusion: The Tcombination of ethanol extracts from Arumanis mango leaves and guava leaves at a dose of 45 mg/200g BW : 12.5 mg/200g BW has the best antidiabetic effect.
