Abstract

Background, Sambiloto (Andrographis paniculata L.) ethanolic leaf extract containing terpenoids was formulated into gel and optimized using Simplex Lattice Design method to enhance comfort and effectiveness of application. Objective, To optimize and evaluate the physical properties of sambiloto (Andrographis paniculata L.) ethanolic leaf extract gel formulation meeting physical quality requirements. Method, This experimental study employed Simplex Lattice Design (SLD) using Design Expert 13 software. Formula optimization varied Carbopol 940 and glycerin concentrations, followed by formulation evaluation and statistical analysis using SPSS version 25. Results, SLD method generated 8 runs for gel formulation design. Runs 1, 2, 4, 6, 7, and 8 met response values for spreadability, adhesiveness, and viscosity parameters. Runs 1, 3, and 5 failed to meet spreadability requirements. Optimization yielded optimal concentrations of Carbopol 940 (0.5%) and glycerin (8%). Statistical analysis showed no significant difference between experimental and SLD predicted values (p>0.05). The optimal gel formulation met organoleptic, homogeneity, pH, and stability testing requirements. Conclusion, The concentration ratio of Carbopol 940 as gelling agent and glycerin as humectant produced sambiloto (Andrographis paniculata L.) ethanolic leaf extract gel meeting physical quality requirements. Recommendation, Future researchers are advised to conduct irritation testing on sambiloto ethanolic leaf extract gel formulations
