Abstract

Background, Ganitri leaves contain flavonoids which have the potential as natural antioxidants. Flavonoids have poor solubility and bioavailability, so nanoparticle technology to be applied. Optimization of nanoparticles to produce quality formulas that can be used by design experts 13. Objective, Determine the optimal formula for ethanol-aquadest extract nanoparticles of ganitri leaves and the characterization. Methods, Flavonoids content was carried out by qualitative and quantitative. Formula optimization used the Simplex Lattice Design (SLD) method with variations of chitosan and Na-TPP based on percent transmittance and trapping efficiency responses. The optimal formula was determined numerically and validated by T-test. Characterization of nanoparticle are organoleptic, pH, particle size, PDI, zeta potential, stability, and total flavonoid of the preparation. Results, The total flavonoid extract were 7,7 mg QE/g, Chitosan 0,3%, and Na-TPP 0,5% is the optimal formula with percent transmittance 97,534±0,8400 and trapping efficiency 92,54±2,0182, where the desirability value is 1. The T-test results show that there is no difference between prediction and testing (p>0.05). The characterization of nanoparticles has a clear appearance, pH 4,39±0,3386; particle size 317±21,7025 nm; PDI 0,652±0,1471; zeta potential 21,13±1,4295 mV; the preparation is organoleptically stable but not zeta potential; and the total flavonoid was 11,624 mg QE/g. Conclusion, A concentration of 0,3% chitosan and 0,5% Na-TPP can produce an optimal nanoparticle formula for ethanol-aquadest of ganitri leaves with characteristics that meet quality requirements. Recommendation, It is necessary to reduce the concentration of Na-TPP and test the antioxidant activity on the nanoparticle preparation of ethanol-aquadest extract from ganitri leaves.
